Studies On Gene Expression Profiles Of Rabbit Retina After Negative Pressure Suction With Microarrays

Purpose: To detect the changes of gene expression profiles of retina by way of gene chip technology methods, and to investigate possible molecular pathogenesis of retinal ganglion cells apoptosis caused by transient high intraocular pressure.

Methods: 12 New Zealand white rabbits were randomized to experimental control and negative pressure suction 3min groups. The experimental control group was treated only by laser, and the experimental group received suction for 3min by negative pressure generator, then the total RNA of retinal tissue was extracted at different recovery stages to detect gene expression profiles by the way of Agilent rabbit one-way gene chip. Comparison was made with control group.

Results: Instant after negative pressure suction, the number of differentially expressed gene was 704.Among them, the 485 genes were upregulated and 219 genes were downregulated. The higher expression of apoptosis genes were CRYAA、CRYAB、TLR3 and KRT18. After 7d in negative pressure suction, the number of differentially expressed gene was 482. Among them, the 178 genes were upregulated and 304 genes were downregulated. The higher expression of apoptosis genes were CRYAB、IL1-BETA and IL1R1. 10d after negative pressure suction, the number of differentially expressed gene was 402. The 213 genes Of these were upregulated and 189 genes were downregulated. The higher expression of apoptosis genes were CRYAB、CRYBA3、CRYBB2、IL1-BETA and IL1R1.

Conclusions: Negative pressure suction for long times(for example 3min), the gene expression had changed. The genes with higher fold change served to protect RGCs from apoptosis.
