The effect of VitE to the Human Retinal Pigment Epithelium Cell Replicative Senesces Stimulated by Blue Light

Purpose :
To observe the effect of VitE to the beta-galactosidae£¨SA-¦Â-Gal£©of cultured human RPE cells replicative senescence stimulated by blue light with fixed wave length and intensity. Then provide the relative theory base of cytobiology pathogenesis of blue light and replicative senescence in AMD and the possible theraphy methord.

Methord:
1.Set up the modle of human RPE cells replicative senescence stimulated by blue light.
Use the fresh human eyeball from accidental death for primary culture. The cell was identified with immunohistochemistry.
2.The usage of VitE and blue light
Divid the cultured human RPE cells into three groups from the third degeneration with different VitE concentration added into the culture fluid: 10¦Ìmol/L,50¦Ìmol/L,100¦Ìmol/L, the control groups are the cells with blue light but no VitE and the cells without VitE and blue light. The blue light with wave length 450nm to 500nm is used, and the light intensity of the cell surface is£¨2000¡À500£©lx. At last check the SA-¦Â-Gal and cell proliferation activity.

Result:
The number of the positive cells of SA-¦Â-Gal dyeing increased with the cell generation, and the proliferation ability reduced. With the VitE added, the positive cell number reduced with the VitE concentration increased, and cell proliferation ability also increased.

Conclusion:
Human RPE cells replicative senescence can be stimulated by blue light, replicative senescence is related to the cell generation, antioxidant like VitE can inhibit the replicative senescence. This may be help for the AMD pathogenesy.
