The experimental application of biological membrane that soaked in 5-FU in glaucoma operation

Observe antiproliferative role of biological membrane that soak 5-FU in experimental rabbit after glaucoma filtration surgery .
Methods 20 (40 eyes) in New Zealand big health rabbits were injected 160u capsular suppot alpha mi protease that establish high intraocular pressure model. Randomly selected 10 rabbits did classic trabeculectomy (trabeculectomy group), the other eye did resection plus single-layer biological trabecular amniotic membrane implantation (membrane group), Another 10 at rabbits did trabeculectomy joint in the scleral flap that placed below 0.25 mg/mL MMC of cotton piece 3min (MMC group), trabecular resection plus other eyes soak 5 - FU solution of biological membrane implantation (5 - FU group). Observe the cornea endothelial cells and quantity, retinal photography, OCT, respectively, after surgery 1d,3d, 14d, 21d 7d, 28d , air embolism method, using immunohistochemical method applied surrounding tissues of rabbit filtering bleb operation ,testing each district organization transformation in transforming growth factor (for TGF - beta 1), observe operation pathological changes.
Results :There is no difference in OCT, retinal photography,endothelial cell count and form of all the observation groups;after three weeks,the start of inflammatory cells in 5 - FU group (average 32.47 ±6.38 / high magnification, vision after three weeks ) less than other groups , TGF beta - 1 expression showed in 5 -FU group less than the rest of groups in the same time(P < 0.05).
Conclusion: Biological membrane that soaked 5-FU applicated in trabeculectomy of lagophthalmos can restrain tissue proliferation of filtration channel ,and high security role.
