柚皮素防治激光诱发大鼠脉络膜新生血管形成

目的:观察柚皮素对脉络膜新生血管形成的防治作用。方法:用彩色微球技术测定柚皮素对高眼压新西兰白兔眼脉络膜血流的影响;用眼电生理检测柚皮素对Long-Evans大鼠视网膜中央动脉结扎后视网膜功能恢复情况;用激光制作Brown-Norway大鼠脉络膜新生血管(CNV)模型,通过眼底荧光血管造影评价柚皮素对CNV的防治作用。结果:柚皮素能改善高眼压兔眼脉络膜血流;促进视网膜中央动脉结扎后Long-Evans大鼠视网膜功能恢复;对激光诱发的Brown-Norway大鼠脉络膜新生血管有抑制作用。结论:柚皮素能显著增加眼部血流,尤其是视网膜和脉络膜,并有抑制激光诱发Brown-Norway大鼠脉络膜新生血管形成的作用。临床研究证明柚皮素还具有较强的抗炎作用。AMD继发的CNV是AMD患者视力丧失的主要原因,现有的研究显示,CNV形成过程中有脉络膜血流受损及炎症机制参与。柚皮素能增加脉络膜血流,也有抗炎作用,有望成为一种防治AMD的药物。 Purpose: To observe the effect of naringenin on choroidal neovascularization in rats. Methods: Colored microsphere technique was used to determine the effect of naringenin on choroidal blood flow in ocular hypertensive New Zealand white rabbits; Electroretinography was used to measure the effect of naringenin on the retinal function recovery after retinal artery occlusion in Long-Evans rats ; Brown-Norway rat eyes were treated with photocoagulation to induced choroidal neovascularization, fluorescein angiography was used to determine the effect of naringenin on choroidal neovascularization in rats. Results: Naringenin increased choroidal blood flow, enhanced the retinal function recovery after retinal artery occlusion , inhibited choroidal neovascularization. Conclusion: Naringenin significantly increase ocular blood flow, especially in retina and choroid. It also showed the effects to inhibited choroidal neovascularization in rats induced by laser. Clinically it was proved that naringenin is a stronger anti-inflammatory drug. Choroidal neovascularization secondary to AMD is the leading cause of blindness in patients with AMD. It have been reported that inflammation and impairment of choroidal blood flow involve the choroidal neovascularization. It were found that naringenin showed increase of choroidal blood flow and effect of anti-inflammation, thus it could be developed into useful drug for prevention and/or treatment of AMD.
