Effect of tTG AS-ODN on the expression of fibronectin in bovine trabecular meshwork cells |

[Abstract] Objective: To observe the effects of antisense oligonucleotides against tissue transglutaminase(tTG) on the expression of fibronectin(FN) in bovine trabecular meshwork cells(BTMC). Methods: After lipofectemine 2000 reagent delivery, BTMC were exposed to 0.3μmol•L-1antisense tTG, serum-free DMEM respectively. After 24 hours later, Immunohistochemistry staining SP Method was used to detecting the effect of antisense tTG on the expression of FN. At the same time, total cell extracts were subjected to western blot and RT-PCR. Results: In comparison with control group, tTG AS-ODN1 and tTG AS-ODN2 could inhibit the FN expression in BTMC at mRNA and protein level(P<0.05), there was no significantly difference between the group of tTG AS-ODN1 and the group of tTG AS-ODN2(P>0.05). Conclusion: It was further confirmed that tTG could promote the expression of extracellular matrix in BTMC. Antisense tTG may be expected to treat primary open angle glaucoma at gene level through inhibition of the expression of FN and other extracellular matrix. |
