Effect of transforming growth factor-β on the expression of connective tissue growth factor in bovine lens epithelial cells

[Abstract] Objective: To observe the effects of transforming growth factor-β(TGF-β)on the expression of connective tissue growth factor(CTGF) in bovine lens epithelial cells(BLECs). Methods: Cultured 2-3 passage BLECs were divided into control group and experimental group with 0×103ng•L-1(control)、0.32×103ng•L-1、1.00×103ng•L-1、3.20×103ng•L-1 TGF-βfor 24h, the mRNA and protein of CTGF in BLEC were examined respectively by semiquantitative RT-PCR and Western blotting. Results: In comparison with control group, the different concentration of TGF-βcould promote the expression of CTGF at mRNA and protein level in BLECs. The result of RT-PCR indicate that there was no significantly difference between the 0.32×103ng•L-1 group and control group(P>0.05), but there were significantly difference between the 1.00×103ng•L-1、3.20×103ng•L-1 group and control group(P<0.05). The result of Western blotting was conformed with the RT-PCR. Conclusion: the data suggest that CTGF play an important role in lens epithelial cells migration, fibrosis and deposition of ECM in posterior capsule opacification.
