一氧化氮及诱导型一氧化氮合酶与二硫化碳所致视网膜功能损害的相关性 |

摘要:目的 探讨一氧化氮(NO)及诱导型一氧化氮合酶(iNOS)与二硫化碳(CS2)所致视网膜功能损害的关系。 方法 将48只SD雄性大鼠随机分为对照组、低剂量CS2染毒组和高剂量CS2染毒组,染毒结束后观察大鼠视网膜电图(ERG)b波振幅及光学显微镜下视网膜结构变化;分光光度法测定NO含量;免疫组织化学法检测诱导型iNOS的表达。 结果 (1)染毒组ERG 的b波振幅均下降,与对照组相比差异有统计学意义(P﹤0.05),高剂量染毒组与低剂量染毒组之间差异亦有统计学意义(P﹤0.05)。(2)染毒组视网膜NO生成增加,与对照组相比差异有统计学意义(P﹤0.01或P﹤0.05),高剂量染毒组与低剂量染毒组之间差异亦有统计学意义(P﹤0.05)。(3)染毒组视网膜iNOS表达增加,与对照组相比差异有统计学意义(P﹤0.01或P﹤0.05);高剂量染毒组与低剂量染毒组之间差异亦有统计学意义(P﹤0.05)。(4)低剂量染毒组NO、iNOS分别与ERG的b波振幅呈负相关(r =-0.674,P﹤0.05;r =-0.701,P﹤0.05);高剂量染毒组NO、iNOS亦分别与ERG的b波振幅呈负相关(r =-0.773,P﹤0.05;r =-0.781,P﹤0.05)。 结论 NO及iNOS与CS2所致视网膜功能损害有关。
Correlation between the level of nitric oxide and inducible nitric oxide synthase of the retina and damage of retina function due to carbon disulfide
Abstract: Objective To investigate the correlation between the level of nitric oxide(NO) and inducible nitric oxide synthase(iNOS) of the retina and damage of retina function due to carbon disulfide(CS2). Methods Forty-eight male SD rats were randomly divided into 3 groups— control group, group received high-dose CS2 and group received low-dose CS2. After treatment, amplitude of b-wave of electroretinogram(ERG) was observed and the histological changes were detected by light microscpy. The content of NO were measured by spectrophotometer. The expression of inducible nitrogen oxide synthase(iNOS) was detected by immunohistochemistry. Results (1) The amplitudes of ERG b-wave reduced in groups received CS2. There were significant difference between control group and groups received CS2(P﹤0.05). There was also significant difference between group received high-dose CS2 and group received low-dose CS2(P﹤0.05).(2)The content of NO increased in groups received CS2. There were significant difference between control group and groups received CS2(P﹤0.01 or P﹤0.05). There was also significant difference between group received high-dose CS2 and group received low-dose CS2(P﹤0.05).(3)The expression of iNOS increased in groups received CS2. There were significant difference between control group and groups received CS2(P﹤0.01 or P﹤0.05). There was also significant difference between group received high-dose CS2 and group received low-dose CS2(P﹤0.05).(4)Negative correlation was found between the level of NO and iNOS of the retina and the amplitude of b-wave of ERG in group received low-dose CS2 (r =-0.674,P﹤0.05;r =-0.701,P﹤0.05). Negative correlation also was found between the level of NO and iNOS of the retina and the amplitude of b-wave of ERG in group received high-dose CS2 (r =-0.773,P﹤0.05;r =-0.781,P﹤0.05). Conclusion NO and iNOS might be involved in the damage of retina function due to CS2.
