摘要
目的:研究Notch信号在RCS鼠视网膜中的发育性表达,探讨Notch信号在损伤的成体内视网膜干细胞增殖分化的抑制机制。
方法:取1、2、4、8周RCS鼠及同期对照鼠的视网膜(每周龄6只),做免疫荧光组织化学检测,分析各视网膜神经元和神经胶质细胞标记物以及 notch信号系统各抗体在视网膜的表达。
结果:GFAP在各周龄RCS鼠及对照鼠均呈阳性表达;Recoverin在RCS鼠1、2周表达阳性,4周表达减弱,8周时没有看到阳性表达,在对照鼠各周龄持续表达;Nestin在各正常组及变性1、2周大鼠视网膜各层免疫荧光检测均为阴性,4、8周视网膜光感受器细胞层部位免疫荧光阳性;Notch1、Hes1在RCS鼠中1、2、4、8周持续呈阳性表达,且8周时表达较强,而在对照鼠中1、2、4中阳性表达,8周时未见表达;NeuroD在1、2、4周龄逐步表达增强,8周弱阳性表达;Mash在1、2、4周龄弱阳性表达,8周未见表达。
结论: notch通过激活 bHLH基因家族的抑制型基因(Hes1)促进了损伤的成体鼠视网膜中内源性干细胞的增殖。

ABSTRACT
Purposes To investigate the inhibition of notch signaling to the differentiation of adult retinal stem cells from which was damaged.
Methods:The RCS rats with inherited retinal dystrophy were divided into 4 groups:1、2、4、8w ( n = 6 animals in each group). These were compared with RCS-rdy + p + rats without retinal degeneration ( control groups, C1w, C 2w, C 4w,C8w). One retina of every animal was prepared for retinal slices and fluorescence histochemistry。Analysis the markers of neuronal cells and glial cells,and the expression of notch signaling in the chronicly injured retina.
Results :GFAP was expressed in the control group s and in the RCS rats;Recoverin was expressed in the RCS rats of 1 and 2week, while the positive exp ression was also observed in the control rats of 1、2、4 and 8week;The positive expression of Nestin was observed in the photorecep tor layers of the 4 and 8week, while all of the retinal layers in the control group s and in the RCS of 1 and 2week rats did not express.Notch and Hes1were expressed in the RCS rats of 1、2、4 and 8 week,and prolonged in 8 week,but were not investigated in the control rats of 8week.NeuroD was expressed in 1and 2 week, and reach the ultimate in 4 week,then become weak in 8 week. Mash1 was weakly expressed in the RCS and control rats ,and was not examined in 8 week.
Conclusions: notch can promote the adult rat retinal stem cells to proliferation by activating the activated bHLH gene Hes1.
