碘化纳切除视网膜色素上皮的鼠模型研究

目的 鼠静脉内或视网膜下腔注射碘化钠建立视网膜色素上皮切除的动物模型。方法 18 只Lewis 鼠36眼,分为四个实验组和两个对照组, 每组3 只鼠。第一组和第二组经尾静脉注入0.08M, pH 7.4碘酸钠120mg/kg。第三组和第四组做视网膜下腔注射碘酸钠150-200µg /每眼(5-7µl),两个对照组分别经静脉和视网膜下腔注入同等量的生理盐水。注射后分别在24小时和1周摘除眼球做光镜、透射电镜和扫描电镜检查。结果 静脉和视网膜下腔注射碘酸钠后24小时,视网膜色素上皮细胞显示脱色素、肿胀或色素增生,并可见斑点状色素上皮脱失;注射后1周可见大面积的色素上皮脱失和玻璃膜裸露,其上的光感受器细胞和其下的脉络膜变形。对照组眼组织显示正常。结论 静脉和视网膜下腔注射碘酸钠后1周可建立鼠视网膜色素上皮切除的动膜模型,它可用于在裸露的玻璃膜上进行视网膜色素上皮移植的研究。 【关键词】视网膜色素上皮;切除; 模型 Na Iodate Model of RPE Debridement in the Rat. Methods A total of 36 eyes from 18 Lewis rats were studied. Groups 1&2 received 120 mg/kg of Na Iodate via the tail vein (iv); groups 3 & 4 received 150-200µg /rat of Na Iodate via the subretinal space (srs) in 5-7 µl. Control groups received a similar volume of normal saline. Eyes were enucleated at 24 hrs and 1 week after injection. Light microscopy, transmission electronic microscopy (TEM), and scanning electronic microscopy (SEM) were performed. Result At 24 hours the eyes of both iv and srs Na Iodate-injected recipients showed plump, densely pigmented and depigmented RPE cells, with patchy areas of RPE loss. At 1 week large areas of RPE loss with bare Bruch’s membrane were observed in both groups injected with Na Iodate. Abnormalities were noted in both the overlying photoreceptors and underlying choroids. Similar changes were not observed with normal saline. Conclusion At 1 week after injection of Na Iodate a model of RPE debridement of Bruch’s membrane in the rat is created after either iv or srs injection. This animal model will allow the study of RPE transplantation on a bare Bruch’s membrane. 【Key Words】Retinal pigment epithelium;Debridement;Model
