Analysis of Id-2 expression uncovers relationship with vasculogenic mimicry in aggressive uveal melanomas

Purpose: To investigate the association between Id-2 expression and clinicopathologic characteristics or tumour vasculogenic mimicry in human uveal melanoma tissues and cells. Methods: 82 cases of paraffin-embedded tissue sections underwent immunohistochemistry staining to examine Id-2 expression and also Periodic Acid Schife (PAS) staining to determine vasculogenic mimicry. Id-2 expressions in highly aggressive uveal melanoma cell lines (Mum-2B) and poorly aggressive cell lines (Mum-2c) (derived from the same patient) were analyzed by Western blot and immunocytochemistry. Results: Out of a total 78 cases, Id-2 expression was found decreased vs 4 cases of nomal tissues. And the down-regulated expression of Id-2 was significantly associated with vasculogenic mimicry( P< 0.01 ). In further analysis of subgroups showing tumours with lowest (scores 2 and 3 ) Id-2 expression showed significantly more chances of vasculogenic mimicry than tumours with lower (scores 4 and 5 ) Id-2 expression ( P< 0.01 ).Analysis of Id-2 expressions in poorly aggressive vs. highly aggressive human uveal melanoma cells by using Western blot and immunocytochemistry confirmed the expression of Id-2 was down-regulated in human melanoma cells. Conclusions: We provide the first evidence to clarify the association between Id-2 expression and intratumoural vasculogenic mimicry which associated with tumour angiogenesis in human uveal melanoma and suggest that Id-2 protein may be an important new target molecule for antiangiogenic drugs in human uveal melanoma therapy.
