An Investigation for Vascular Mimicry in Optic Nerve Glioma by combined histochemistry stain

Abstract Purpose: To study the relationship between altered blood vessel stucture and vascular mimicry formation in optic nerve glioma(ONG). Methods: Twenty five specimens were harvested from patients undergoing ONG excision. They were studied under the light microscope by HE, PAS histochemistry, immunohistochemistry and immunohistochemistry combined with PAS histochemistry double stain. Results: Vascular mimicry was identified in all the ONG cases. Some loose reticulate region,stream-like channels and large cystilike cavities were found in the ONG tissue sections, protein deposits and red blood cells existed within the entocoele of these strctures. The protein deposits and septas of meshwork were PAS-positive. The meshwork was composed of delicate projections extended from GFAP-positive tumor cells and PAS-positive extracelluar matrices (ECM); some slender and branching projections encircled by extracelluar matrices extended from GFAP (Glial Fibrillary Acidic Protein) positive tumor cells, anastomosing to form meshwork. The capillary walls were surrounded or half surrounded by some PAS-positive ECM which were not uniform in thickness. Conclusions: Several types of vascular mimicry were identified in ONG. The pathological changes of blood vessel walls render the leakage of plasma from blood vessels into adjacent interstitium, and many asteroid processes, plenty of extracelluar interspace might be the morphological and biological foundation for the formation of vascular mimicry.
