Preliminary study on effect of distance transportation on mid-term medium preserved rabbit corneal endothelium

Objective:To explore the effect of mechanical agitation on corneal endothelial function. Methods:36 corneas of New Zealand rabbits were obtained and preserved in DX mid-term medium for 7 days. Laboratory shaker was adopted to simulate the transportation condition. 6 corneas were used as control and they were all dyed directly after preservation and others were grouping according to 2 cues. First, 3 groups (6 corneas in each group) were set up according to different transportation time with the max vibration intensity as primary standard. Second, 3 groups (6 corneas in each group) were set up according to different vibration intensity with the max transportation time as primary standard. All corneal buttons were examined by trypan blue combined with alizarin red staining and calculate the CEC density, the viability rate and percentage of hexagonal cells. Na+-K+ ATPase and SDH enzymohistochemical staining and image analysis system were employed to calculate and analyze average IOD of two enzymes in CEC from each group. Sample from the most intense and the longest time vibration group was examined by transmission electron microscopy. Compared with the control, the ultrastructural changes of CEC were observed. Results:Under laboratory simulated transportation state, transportation effectiveness was not affected significantly with designed transportation time and designed transportation intensity. The viability of endothelium from control group and experimental groups were all having the significant higher vital rate and cell shape were fundamental normal. Expression of two enzymes has no significant difference among groups. By TEM observation, CEC from experimental group and control group were all continuous. Cell junctions were tight and cytoplasmic organoids were very abundant. Conclusion:DX medium could be used as a safe and reliable medium for long-distance shipment which can maintain corneal vitality and fit the requirement of PKP.
