人类脐血干细胞对大鼠外伤性视神经损伤视觉电生理的影响

目的 观察人类脐血干细胞的移植对SD大鼠外伤性视神经损伤后视觉诱发电位 (F-VEP)的影响。
方法 健康成年SD大鼠随机分为视神经钳夹30S组及60S组,所有大鼠的左眼为损伤眼,右眼为正常对照眼。两组按视神经损伤后处理方式的不同随机分为对照组(A组)和治疗组(B、C、D组),A组不予以药物治疗,治疗组分别予以玻璃体腔内注射神经营养因子(B组)、脐血干细胞(C组)、脐血干细胞+神经营养因子混合液(D组)。所有大鼠均在损伤后1小时、第7天、第14天、第21天和第28天进行损伤眼和正常对照眼的F-VEP检查,记录波幅及峰潜时,并进行统计分析。
结果  1. F-VEP的波幅:视神经钳夹30S组,D30组和A30组波幅分别为6.04±1.3、4.56±0.76; 钳夹60S组,D60组波幅为5.89±1.1,A60组为4.41±0.56,差异有显著性 (P<0.01)。2. F-VEP的P1峰潜时:视神经钳夹30S组,D30组P1峰潜时为89.5±1.6,A30组为100.6±3.3;钳夹60S组,D60组为84.5±1.6,A60组为95.5±3.1,差异有显著性(P<0.01)。 
结论 人类脐血干细胞移植可以有效促进视神经损伤后神经传导功能的恢复,用神经营养因子基因修饰后移植,能进一步提高上述功能。

Objective  To investigate whether transplantion of human umbilical cord blood stem cell has the effect on flash visual evoked potential (F -VEP) of rats which were treated with the traumatic optic nerve injury,
Methods  96 healthy SD rats were randomly divided into 2 groups (n=48) according to the optic nerve clamping time(30 second group and 60 second group). The left eyes in all groups were clamped and the right eye was served as control. Then, these two groups were randomly divided into 4 subgroups (n=12) with the difference treatment after optic nerve clamping. Group A was control which was no treatment after injury. Group B to D were treated with human umbilical cord blood stem cells, neurotrophic factors, or the mixture respectively by vitreous cavity injection. And the F-VEP was recorded in all rats at 1 hour, 1, 2, 3 and 4 week day after clamping. The statistical analysis was done in wave amplitude and wave latency.
Results  In the 30S injury group, the wave amplitude of group D and A was 6.04 ± 1.3, was and 4.56 ± 0.76 respectively. In the 60S injury group,it was 5.89 ± 1.1 and 4.41 ± 0.56 in group D and A. There was significant difference.(p<0.01). In the 30S injury group, the wave latency in group D and A was 89.5±1.6 and 100.6±3.3 respectively. In the 60S injury group, it was 84.5±1.6 and 95.5±3.1 in group D and A. There was significant difference.(p<0.01).
Conclusion  The transplantation of human umbilical cord blood stem cell after the traumatic optic nerve injury can improve the recovery of nerve impaulse transition, which was more effective on nerve recovery by combined with neurotrophic factor.
