全反式维甲酸对婴幼儿晶体上皮细胞增殖的抑制作用

目的 研究在体外细胞培养条件下,全反式维甲酸对婴幼儿晶体上皮细胞增殖的抑制作用及有效抑制浓度,为预防婴幼儿后囊膜混浊发生的研究提供线索。方法 传代培养的婴幼儿晶体上皮细胞经计数后在370C,5% CO2的条件下培养48 小时,再加入浓度分别为2、5、10、15、20μg/ml全反式维甲酸完全培养基作为实验组,继续培养24及72小时后进行活细胞计数并与所设空白对照组对比,观察全反式维甲酸对传代细胞的抑制作用并找出有效作用浓度及半效抑制量(ID50)。结果 全反式维甲酸对体外培养的婴幼儿晶体上皮细胞的增殖有明显的抑制作用:从2μg/ml浓度即有显著的抑制效应,到10μg/ml浓度时达到最大作用。全反式维甲酸作用24小时的ID50是9.22μg/ml;作用72 小时的ID50为4.75μg/ml。结论 全反式维甲酸能有效地抑制体外培养的婴幼儿晶体上皮细胞的增殖,有可能成为预防婴幼儿晶体后囊膜混浊的理想药物。 关键词:全反式维甲酸; 后囊膜混浊;婴幼儿晶体上皮细胞;ID50 The inhibition of the infant lens epithelial cell proliferation by Retinoic Acid in Vitro SHI Lei. Department of Five organ, Xingcheng city hospital, Liaoning Province Abstract Objective To investigate the inhibitory activity and the effective concentration of all-trans retinoic acid against the proliferation of infant human lens epithelial cells (HLEC) in vitro, which give a clue to the study on the prevention of infant posterior capsule opacification. Methods The passage cultured infant HLEC were incubated at the condition of 370C, 5% CO2 for 48 hours after counted and then exchanged with the medium contained 2,5,10,15,20μg/ml of all trans retinoic acid respectively as the tested groups, which were compared with the blank control group with normal medium. Then these passage cells were counted again after another 24 and 72 hours to analyze the inhibitory function by various concentration all-trans retinoic acid and found out the its effective concentration as well as half inhibitory dose (ID50). Results There is a prominent inhibition on the proliferation of infant HLEC by all-trans retinoic acid: from 2 μg/ml until 10μg/m to reach its peak, the 24 hours ID50 is 9.22μg/ml and the 72 hours one is 4.75μg/ml. Conclusion All-trans retinoic acid can inhibit the cultured infant HLEC proliferation in vitro and may become an ideal agent in preventing the infant posterior capsule opacification.
