Expression and correlation of ILK and downstream molecules in RF/6A cells under the hypoxic condition

Objective  To investigate the expression of ILK , HIF-1α ,SDF-1 and ICAM-1 in RF6A cells under the hypoxic condition and the relationship between ILK and HIF-1α ,SDF-1 and ICAM-1.
Methods  Cultured RF6A cells were maintained with Cocl2 for 0h6h12h24h48h72h respectively in vitro. The expression of ILK, HIF-1α,SDF-1 and ICAM-1 were quantified by Real Time PCR and Western blot. To quantify the expression of ILK, HIF-1α,SDF-1 and ICAM-1 in RF6A cells under the hypoxic condition after the activity of ILK was inhibited with LY294002.
Results Under hypoxia conditionILK , HIF-1α ,SDF-1 and ICAM-1 co-expressed in RF6A cells. The expression levels of the four factors were all ascended and reached the summit after 12~24h by hypoxia while descended after 72hHowever, this upward trend was inhibited after the activity of ILK was inhibited .
Conclusion  Hypoxia can promote the expression of ILK , HIF -1α , SDF -1 and ICAM -1 in RF/6A cells. The expression of ILK and HIF - 1α , SDF -1 and ICAM -1 was positively correlated .
