FDM小鸡模型巩膜中MMP-2的测定

FDM小鸡模型巩膜中MMP-2的测定 【中文摘要】  目的 通过建立动物的近视眼模型,观察近视眼发展期和恢复期屈光状态的改变以及基质金属蛋白酶MMP-2的含量变化,探讨小鸡近视眼模型的巩膜重塑过程中的生化学变化机制。 方法 实验动物为2天龄SPF美国进口白种莱航鸡,分成四组,用透明眼罩遮盖试验组(A组和B组)动物的右眼进行单眼形觉视觉剥夺,对侧眼为配对对照组,并设立正常对照组(C组和D组),遮盖后21天检测双眼屈光状态和眼轴,处死A组和C组动物,分离7mm直径的后巩膜组织,保存于-80℃的低温冰箱;继续饲养B组(去遮盖)和D组,去遮盖后21天检测双眼屈光状态和眼轴,处死后分离7mm直径的后巩膜组织,用酶活性分析法(activity assay)测定所有动物巩膜中MMP-2的含量,并将实验组和对照组的数据进行统计学分析。 结果 遮盖21天A+B组左右眼屈光度,有显著性差异,平均差值-28.38+6.72D;A组左右眼眼轴,有显著性差异,平均差值1.47+0.37mm;遮盖眼后极部巩膜中的MMP-2含量显著性升高,A组遮盖眼与无遮盖眼间配对t检验,t=16.152,P<0.001,有显著性差异,平均增加76.00%+15.20%。去遮盖21天B组左右眼屈光度,无显著性差异,平均差值0.19+0.92D; B组左右眼眼轴,无显著性差异,平均差值0.15+0.50mm;去遮盖眼后极部巩膜中的MMP-2含量明显下降,ANOVA方差分析,各组间F=0.243,P=0.786,无显著性差异。 结论 研究数据表明形觉剥夺性近视眼的小鸡模型中,近视眼的形成与后巩膜中的MMP-2水平有联系,而且MMP-2的含量在近视眼发展期和恢复期存在一定的变化规律。 【关键词】 近视眼;形觉剥夺;巩膜;基质金属蛋白酶;MMP-2 The Content Of MMP-2 In The Sclera Of Myopic Chick Eyes 【Abstract】 Purpose To study the biochemical change of the sclera of FDM chicks during the process of sclera remodeling, and asses the role of this metalloproteinase MMP-2 in ocular elongation associated with experimental myopia and the dynamic change of it. Method 32 2-day-old white Leghorn chicks were divided into four groups. The chicks were raised under the environment of stable temperature and moisture, 12-hour light-12-hour dark cycle and were checked daily. Form-deprivation myopia was induced in 2-day-old white Leghorn chicks by applying translucent plastic goggles attached on the right eyes—A and B group. Untreated fellow eyes served as paired controls, and also normal controls—C and D group— were set. The goggles remained in place for 21 days, and all the eyes were checked up for refractive condition. Then killed the chicks of A and C group, and the sclera punches (7 mm in diameter) were excised from the posterior poles of control and form-deprived eyes by surgical trephine after measuring the axial of the eyes. Keep the samples in the -80℃ refrigerator. Half of the treated chicks—B group— were raised for more 21 days. All the eyes were checked up for refractive condition and axial length. The sclera punches (7 mm in diameter) were also excised from the posterior poles. The amount of MMP-2 was measured by activity assay and the data were analyzed by SSPS 11.0. Result Significant differences were detected in refraction and eye elongation of the covered eye and no significant differences were found after recovery from FDM between treated eye and normal control. Significant differences were detected in MMP-2 activity between normal and deprived posterior sclera when treated with APMA. The amount increased 76.00%+15.20% after form-deprived for 21 days and decreased near normal level when the treated eyes recovered for 21 days. Conclusions These data indicate that form-deprived myopia is associated with an increased amount of MMP-2. And the amount of MMP-2 in the posterior sclera has some sort of regular change during the development and recovery of form-deprived myopia. 【Key Words】 myopia; form-deprived; sclera; metalloproteinase; MMP-2
