[摘要] 目的 检测培养的眼眶成纤维细胞(OF)是否存在表达CD90抗原的差异,是否存在CD90+和CD90-两种亚型,比较甲状腺相关眼病(TAO)患者和正常对照眼眶不同部位CD90+细胞的比例,为研究眼眶成纤维细胞潜在的功能异质性奠定基础。方法 培养TAO病人和正常对照眼眶成纤维细胞,流式细胞术检测CD90的表达。结果 正常人、TAO患者的眼外肌、眼眶脂肪/结缔组织来源的成纤维细胞均有部分细胞表达CD90。正常人/眼外肌来源 OF、TAO患者/眼外肌来源 OF、正常人/眶脂肪来源 OF、TAO患者/眶脂肪来源眼眶成纤维细胞CD90+细胞的比例分别为82.95%±6.49%,80.83%±7.14%,64.55%±4.45%,59.20%±11.19%。眼外肌来源的成纤维细胞中CD90+的细胞比例高于眼眶脂肪/结缔组织来源的成纤维细胞(P﹤0.05),而正常人与患者之间没有显著差异(P﹥0.05)。
【Abstract】 Objective In this report we investigated whether orbital fibroblasts exhibit heterogeneous with respect to surface CD90 expression in culture. Methods Human orbital fibroblasts were cultivated , indirect immunofluorescence and flow cytometric analysis were used to determine the levels of CD90 expression . Results Orbital fibroblasts exhibit a bimodal distribution with regard to the expression and surface display of CD90.The CD90+ cells ratio in fibroblasts from normal extraocular muscles、TAO extraocular muscles、normal connective/adipose tissue、TAO connective/adipose tissue was 82.95%±6.49%,80.83%±7.14%,64.55%±4.45%,59.20%±11.19% respectively.There were more CD90+ fibroblasts in extraocular muscles than in connective/adipose tissue. |
