THE EFFECT OF A NEW AMNIOTIC EXTRACTION ON SD RATS’ ALKALI-BURNED CORNEA

Purpose: to test whether amniotic extration has therapeutic effects on SD rats’ alkali-burned cornea. Methods: prepare amniotic extraction and dilute into 1.0mg/ml, 0.2mg/ml, 0.1mg/ml and 0.05mg/ml. An alkaline burn cornea model was developed in both eyes of SD rats using 1.0 N NaOH. Drip extraction at different concentrations, normal saline and 0.1% dexamethasone 4 times per day for 7 days. Eye evaluation was recorded at day 1, day 2, day 3, day 5 and day 7. At day 7, rats were sacrificed and perfused to get the ratio of angiogenic area to total cornea area (RA). All the data were processed by SPSS11.0. Result: Amniotic extraction exhibited significantly anti-angiogenic and anti-inflammatory effects, especially at concentration of 0.2mg/ml, though less powerful compared with 0.1% dexamethasone. Amniotic extraction significantly promote epithelial healing and reduced scar formation. Conclusion: Amniotic extraction prepared by our method may have integrated therapeutic effects on treating alkaline-burned cornea and provides convenient preservation.
