Abstract:Objective To investigate the effect of transforming growth factor-β(TGF-β) and interferon-γ( IFN-γ)on the immunological competence of human retinal pigment epithelialium cells(HRPE) in vitro. Methods The immunological competence of HRPE was observed by flow cytometry to detect the expressions of HLA-ABC、HLA-DR and ICAM-1. Result The cultured HRPE cells expressed high levels of HLA-ABC and low levels of HLA-DR molecules. After cultured with the above cytokines, IFN-γup-regulated the HRPE’s expression of HLA-ABC, HLA-DR and ICAM-1(P<0.001). TGF-β1 and TGF-β2 could not down-regulate the HRPE’s expression of HLA-ABC, HLA-DR and ICAM-1 without the IFN-γ
stimulation ( P>0.05). But they both suppressed the HRPE’s expression of HLA-DR induced by the IFN-γstimulation ( P<0.001) and TGF-β2 suppressed the HRPE’s expression of HLA-ABC induced by the IFN-γstimulation. (P=0.48). Conclussion IFN-γstrengthens the antigenicity of HRPE, which plays a role in the immunological rejection after HRPE transplantion. TGF-β1 and TGF-β2 weakens that of HRPE, which plays a role in avoiding the immunological rejection after HRPE transplantion.
