| Purpose: To investigate the association of the APOE gene polymorphisms (-491A>T, -427T>C, -219T>G andε2/ε3/ε4) with primary open angle glaucoma (POAG).
Methods: A cohort of totally 142 unrelated Taiwan Chinese POAG patients were examined, including 94 high tension glaucoma (HTG) and 48 normal tension glaucoma (NTG). 77 unrelated subjects served as control subjects also were examined. The ε2/ε3/ε4 polymorphism of APOE gene was genotyped by polymerase chain reaction (PCR) and restriction fragment length polymorphisms (RFLP). The promoter polymorphisms of APOE gene were genotyped by direct sequencing. The genotype and allele frequencies of APOE polymorphisms of patients and control subjects were compared using chi-square test.
Results: There was no significant difference in genotype frequency of APOE exon-4 polymorphisms between HTG patients and control subjects (P=0.179), NTG patients and control subjects (P=0.422), or POAG patients and control subjects (P=0.258). No significant difference in genotype frequencies between POAG cases and controls was found in polymorphisms -491A>T, -427T>C and -219T>G with the p-value of 0.813, 0.663, 0.768, respectively. For the allele frequency of APOE exon-4 polymorphisms, no significant difference was detected, either. The p-value is 0.476, 0.373, 0.391 for HTG/control, NTG/control, POAG/control, respectively. The allele frequencies of polymorphisms -491A>T, -427T>C, -219T>G were not significantly different between POAG group and control group.
Conclusions: According to our findings, the allele and genotype frequencies of APOE gene polymorphisms are not associated with POAG in Taiwan Chinese.
