Abstract
Purpose: Muscarinic acetylcholine receptor (mAChR) antagonists were considered to mediate the effect of slowing the progression of myopia in experimental animals and clinical trials via binding mAChR in eye. The aim of the study was to investigate mAChRs (M1 to M4) expression in retina and sclera in form-deprived myopia (FDM) eyes of guinea pigs and determine whether these two positions are potential sites of action for mAChR antagonists.
Methods: FDM models were established in 15 guinea pigs. Real time RT-PCR was used to detect mRNA expression of mAChRs; immunohistochemistry and Western blot analysis were used to measure the expression of mAChRs proteins in both FDM and open eyes.
Results: mRNA of mAChRs didn’t show significantly different expression between FDM and open eyes both in retina and sclera (P>0.05). Immunohistochemistry and Western blot analysis demonstrated the protein expression of mAChRs in FDM eyes was comparable with that in open eyes.
Conclusions: Findings of this study indicate that both retina and sclera are not potential sites of action for the muscarinic antagonists in preventing myopia. The actions of these drugs are through muscarinic receptors in extraretinal and extrascleral tissues, or nonmuscarinic mechanism, is to be investigated.
