目的 探讨苦参碱(Ma)对体外培养的人视网膜母细胞瘤细胞株的增生抑制及凋亡的影响。
方法 将0.4mmol/L浓度的苦参碱作用于体外培养的人视网膜母细胞瘤细胞株12小时、24小时、48小时,阴性对照组以等量含10%类标准胎牛血清 RPMI-1640 培养液替代苦参碱。观察苦参碱对视网膜母细胞瘤细胞生长增殖的影响,流式细胞仪检测细胞凋亡率。
结果 12h后可观察到细胞开始凋亡,48h内呈时间依赖性。实验组与对照组比较差异具有统计学意义(р<0.05)。苦参碱作用Rb细胞12小时后早期凋亡细胞占凋亡细胞的89.20%,24h早期凋亡细胞占.78.97%,.48h早期凋亡细胞占76.12%。
结论 Ma能够使视网膜母细胞瘤细胞的增殖抑制和凋亡诱导,促进凋亡能力有时间依赖性。
Objective To study the effect of matrine on the apoptosis of retinoblastoma cells.
Methods Retinoblastoma cells was cultured in 0.4 mmol/L matrine for 12,24,48 hours respectively, the control group was cultured in equivalence of the standard the nutrient solution RPMI-1640 at concentrations of 10% fatal bovine serm substitute for matrine. The apoptosis rate was evaluated by flow cytometry.
Results Apoptotic cells was observed after 12h, then developed in a time-dependent manner 48 hours. The difference was statistical significance compared with the control group.(р<0.05). Followed by 12h of matrine treatment, the ratio of viable apoptotic cells to total ones was 89.20%, and then decreased to 78.97% and 76.12% by 24h and 48h respectively.
Conclude Matrine can inhibit the proliferation of retinoblastoma cells though induction of its apoptosis in time-dependent manner. |
