| Characters of newborn human retinal microvascular endothelial cells |

Purpose: To investigate the properties of newborn human retinal microvascular endothelial cells (RMECs) to facilitate studies in vitro .
Methods: Newborn human RMECs were isolated and characterized for expression and localization of endothelial cell markers by immunofluorescence staining of CD31、vWF 、 uptake of acetylated low-density lipoprotein and tube formation. vWF distribution in cells was observed by confocal immunofluorescence microscope and Western blot analysis.
Results: High purity RMECs can be obtained readily and formed capillary-like networks on Matrigel well. vWF of newborn hRMECs mainly located in nuclear, while it distributs in cytoplasm in adult hRMECs from evidenced reports before. .
Conclusions: Human RMECs of newborn have some specific properties. Cells properties related with age of donors should be considered in study in vitro, especially on some age specific disease, such as retinopathy of prematurity. |
