摘要 目的 观察 rhEGF 滴眼液对持续性高眼压致角膜内皮损伤修复的作用。方法 24只本学院动物中心提供的大白兔, 两只眼前房分别注入 1%4000cps甲基纤维素0.20ml形成眼压约30mmHg~40mmHg, 持续约 1~2周的高眼压动物模型。每只兔的双眼随机分为实验组和对照组。实验组滴用40μg/ml的rhEGF (recombinant human epidermal growth factor) 滴眼液 4次/天, 对照组不用药。在选定的时间分别观察角膜内皮细胞密度 (endothelial cell density, ECD);内皮细胞面积变异系数 (coefficient of variation ,CV);六边形细胞出现率 (6A) 及透射电镜 (transmission electron microscope, TEM) 检查。 结果 1. 伤后第一天实验组与对照组角膜内皮细胞密度,六边形细胞出现率及角膜厚度比较无显著性差异。2. 伤后第三天,实验组的 ECD,6A 较对照组高,而 CV 且比对照组低。实验组的角膜厚度比对照组薄。3. 伤后第七天实验组的角膜内皮细胞密度,六边形细胞出现率明显高于对照组。而内皮细胞面积变异系数明显小于对照组 。实验组的角膜厚度显著较对照组低 。4. 伤后第七天透射电镜检查显示 rhEGF 治疗组再生的角膜内皮细胞较对照组可见更多的粗面内质网和核糖体。结论 局部滴用 rhEGF可有效促进持续性高眼压致兔角膜内皮损伤的修复。
Abstract Objective To investigate the influence of rhEGF eyedrops on rabbit corneal endothelial wound healing in vivo. Methods The corneal endothelial wound by the continuous high intraocular pressure were made in 24 rabbits. For each rabbit , one corneal was treated with 40μg/ml rhEGF eyedrops qid, while the mate served as control don’t treat. At selected times after injury, the corneas were evaluated for corneal thickness, the endothelial cell density, coefficient of variation, 6A and TEM. Results
1. One day after injury, the both group indicated the morphological changes and functional disorder. But both group had no significant difference. 2. At 3 day after injury, the ECD, 6A of rhEGF–treated corneas were higher than that of the control group, while the CV of the rhEGF–treated was less than those control group. the corneal thickness of rhEGF–treated group were thinner than that of control group. 3. Seven days after injury, endothelial wounds of eyes treated with rhEGF eyedrops healed more rapidly than the control groups. At seventh day, the corneas of rhEGF–treated group were significantly thinner than those control group. the endothelial cell density,6A of rhEGF–treated corneas was significantly higher than that of control group. But the coefficient of variation of rhEGF–treated corneas were significantly less than those control group. 4. The electron microscope examination showed that regenerated endothelial cells in the rhEGF group were longer, with more rough endoplasmic reticulum and ribosome in them. Conclusion The local applying of rhEGF eyedrops after the corneal endothelial wound by the continuous high intraocular pressure significantly promoted the healing of rabbit corneal endothelial wound. |
