胰岛素对人视网膜色素上皮细胞表达低氧诱导因子-1α的影响

目的 研究胰岛素诱导人视网膜色素上皮细胞(RPE)低氧诱导因子-1α(HIF-1α)的表达, 及川芎嗪对其的影响。方法 应用免疫组化法检测不同浓度胰岛素作用下RPE中HIF-1α的表达、同一浓度胰岛素作用于RPE不同时间HIF-1α的表达、不同浓度川芎嗪对胰岛素 作用下RPE中HIF-1α的表达的影响。结果 胰岛素作用4h,随着胰岛素浓度的增加,RPE中HIF-1α的表达逐渐增加,以8×106 U•L-1最为明显; 8×106 U•L-1胰岛素作用不同时间,以4h时RPE中HIF-1α的表达较高;不同浓度的川芎嗪对胰岛素作用下的RPE中HIF-1α的表达无影响。结论 胰岛素可诱导RPE中HIF-1α的表达,川芎嗪对胰岛素诱导的RPE中HIF-1α的表达无影响。 Objective To study the effects of insulin on the expressing of hypoxia-inducible factor-1α(HIF-1α) in retinal pigment epithelial (RPE).Methods Different concentrations of insulin acted on RPE cells at the same time, the same concentration of insulin acted on RPE at different time, and different concentrations of Tetramethylpraine( TMP) acted on RPE acted by insulin. The expression of HIF-1α were detected by the method of immunohistochemical ,and the picture was analyzed by computer image analysis meter. Results Acted by insulin at 4 hours, with the increasing of concentrations , the expression of HIF-1α on RPE increased gradually, and 8×106 U•L-1 was the most significant. Acted by 8×106 U•L-1 insulin at different time, the expression was the most obvious at the 4 hours stimulated time. Different concentration of TMP had no effect on the expressions of HIF-1α in RPE induced by insulin. Conclusions Insulin induced RPE expressing of HIF-1α,and the TMP can not prevent the effect.
