端粒酶编码区的反义寡核苷酸对人视网膜色素上皮细胞增生的影响
【摘要】目的 探讨不同浓度的端粒酶编码区的反义寡核苷酸(AS-ODN)对人视网膜色素上皮(RPE)细胞增生活性的影响。方法 用不同浓度(0,5,10,15umol/L)的端粒酶编码区的AS-ODN处理人RPE细胞后24小时,使用MTT法测定RPE细胞增生的抑制率,使用流式细胞仪检测其对RPE细胞周期的影响. 结果 不同浓度端粒酶编码区AS-ODN处理过的RPE细胞的抑制率随AS-ODN的浓度增高而升高,并将RPE细胞阻滞在G0/G1期. 结论 端粒酶编码区的AS-ODN对RPE细胞有抑制作用且呈浓度依赖性,这一结论可能成为增生性玻璃体视网膜疾病(PVR)基因治疗的一个新靶点.

Effect of Antisense Oligonucleotid of telomerase on proliferation of human cultured retinal pigment epithelial cells
【Abstract】 Objective To investigate the inhibition of AS-ODN of telomerase on the proliferation of hRPE cells. Methods The cultured hRPE cells were treated with AS-ODN of telomerase at various concentration of 0 umol/L,5 umol/L,10 umol/L,15 umol/L for 24 hours﹒MTT assay was used to detect inhibiting rates of cultured hRPE cells﹒Flow cytometry was used to analyze cell cycle. Results Inhibiting rate of cellular growth was positively correlated with the concentration of AS-ODN of telomerase ,and the percentage of cells in G0/G1 phase increased from42.4%to74.2%,the percentage of cells in G2 and S decreased from36.4%to14.9%and from21.2%to10.9%respectively(P<0.001) Conclusion AS-ODN of telomerase can inhibit the proliferation of hRPE cells thus providing a potential ,novel gene therapeutic approach to PVR prevention.
