Objectives:investigate the variations of concentrations of inflammation cytokines (IL-1、IL-2、IL-6) in the rabbits cornea during the process of healing after creating the flap with femtosecond laser, and compare with that during the same process with mechanical method.
Methods:Utilized the femtosecond laser(intralaser) to create the flap with the thickness of 110 um in the corneas of right eyes of 15 Newzealand rabbits, utilized the microkeratome (Morria M2) to create the flaps in corneas of left eyes of the same rabbis. Measure the concentrations of IL-1、IL-2、IL-6 in the corneas 1 day、1 week、1 month after surgery with method of ELISA.
Results:1 day、1 week、1 month after femtosecod laser ablation, concentrations of IL-1 in the rabbits cornea are 115.17、52.75、196.81 pg/ml respectively. 1 day、1 week、1 month after mechanical ablation, concentrations of IL-1 in the rabbits cornea are 88.29、126.65、356.29 pg/ml respectively. (Table 1 & Figure 1). 1 day after both methods of ablation the concentration of IL-1 in the cornea increased to the same level. During the period of 1 month after mechanical ablation the concentrations of IL-1 increase consecutively. 1 week after femtosecond laser ablation the level of IL-1 in the rabbit cornea decrease to the half of that one day after laser ablation. 1 month after femtosecond laser ablation the concentration of IL-1 was twice of that one day after laser ablation, but which was not beyond the level of that one month after mechanical ablation.
1 day、1 week、1 month after femtosecod laser ablation, concentrations of IL-2 in the rabbits cornea are 0.09、0.25、0.33 ug/L respectively. 1 day、1 week、1 month after mechanical ablation, concentrations of IL-2 in the rabbits cornea are 0.19、0.22、0.36 ug/L respectively (Table 2 & Figure 2).
The level of IL-2 in the rabbits cornea increased consecutively during the period of 1 month after both methods of ablation.
1 day、1 week、1 month after femtosecod laser ablation, concentrations of IL-6 in the rabbits cornea are 5.51、8.18、3.65 pmol/L respectively. 1 day、1 week、1 month after mechanical ablation, concentrations of IL-6 in the rabbits cornea are 11.27、6.18、6.78 pmol/L respectively (Table 3 & Figure 3). One day and one month after femtosecond laser ablation lead to higher level of IL-6 in the rabbit cornea than mechanical ablation.
Conclusion:
Mechanical method resulted higher concentration of IL-1 in the rabbits cornea than the femtosecond laser ablation one month after ablation. The level of IL-2 in the rabbits cornea increased consecutively during the period of 1 month after both methods of ablation. 1 day after ablation mechanical method lead to higher level of IL-2 than femtosecond laser ablation. During the period of one month after surgery femtosecond laser ablation resulted in higher concentration of IL-6 in the rabbit cornea than the mechanical ablation, expect that one week after ablation.
