Age-related macular degeneration (AMD) is the important cause of irreversible central vision loss in elderly people and LOC387715 A69S polymorphism has been found to be associated with AMD. So we pooled the results of all available 9 papers examining the association between LOC387715 A69S genetic variants and AMD to estimate the magnitude of the gene effect and the possible mode of action. Data extraction and study quality assessment were performed in duplicate, and heterogeneity and publication bias were explored. There was strong evidence for association between LOC387715 A69S and AMD, with those having TT and GT genotypes being roughly 7.5 and 2.3 times more likely to have AMD than patients with GG genotype, suggesting a largely co-dominant mode of action. This meta-analysis indicates a multiplicative model with each T allele of LOC387715 A69S increasing the odds of AMD by roughly 2.6-fold. |
