【摘要】目的 为提高眼库对角膜材料的HBV检测提供一种可选择的高灵敏度和特异性的新的筛查方法,并探讨不同眼组织中HBV DNA含量的差异在临床应用中的意义。方法 将10例HBV感染者的取角膜、球结膜及筋膜、色素膜及血清标本提取DNA,使用荧光定量PCR技术检测其单位质量中HBV DNA含量。结果 眼组织中HBV DNA的浓度明显高于血清中HBV DNA的拷贝数;三种组织中,色素膜中HBV DNA的浓度最高;虹膜组织中HBV DNA浓度的变异度最小。结论 荧光定量PCR技术可以作为一种新的方法应用于临床眼库的HBV等病原体的筛查,能够同时检测血液标本和眼组织标本。在难以取得血液标本的情况下,可以选择球外筋膜或者虹膜组织进行HBV DNA的检测。
Detection of HBV DNA in Different Tissue of Eyes and Application
【abstract】objective To provide a new method with high sensitivity and specialty to screen HBV DNA in cornea for eye bank, and discuss the signification of the different content of HBV DNA in different tissues in clinic application. Method DNA was extracted from the samples of cornea, conjunctiva, pigment membrane, and blood serum of 10 HBV infectors. The fluorescent quantitative PCR was used to detect the content of HBV DNA in standard unit of quality. Result The content of HBV DNA in the tissues was significantly higher than blood serum; in those tissues, pigment membrane has the highest content; conjunctiva has the smallest variation of those results. Conclusion Fluorescent quantitative PCR can be a new method used for the screening of HBV DNA in eyes bank, and it can detect not only blood samples, but also tissue samples. In the situation that blood sample is difficult to be acquired, we can choose conjunctiva and its outer tissues for detection.
