大鼠骨髓间充质干细胞体外诱导为光感受器细胞的实验研究 |

Purpose To isolate and culture rat mesenchymal stem cells and induce rat MSCs into photoreceptor cells in-vitro.
Methods The mesenchymal stem cells were isolated and cultured by their adherent ability to the wall of culture dishes. Then the cells were characterized by flow cytometry. The MSCs were induced by murine EGF for 8-10 days in-vitro. Then we used immunohistochemistry staining and flow cytometry to see whether they could express rhodopsin and differentiate into photoreceptor cells.
Result We obtained a lot of MSCs which were highly pure. After induction by EGF, 15.3% MSCs expressed rhodopsin.
Conclusions After induction by EGF, about 15.3% MSCs could differentiate into photoreceptor cells.
目的 分离培养大鼠骨髓间充质干细胞,尝试在体外将其诱导为光感受器细胞。
方法 采用贴壁筛选法分离、培养SD大鼠骨髓MSCs细胞,经流式细胞仪鉴定后,用含小鼠EGF的培养液诱导MSCs细胞8-10天。用免疫组织化学染色观察诱导后的细胞是否表达视紫红质,并用流式细胞术检测表达视紫红质的细胞比例。
结果 采用贴壁筛选法可获得大量高纯度的大鼠骨髓MSCs细胞,经EGF诱导后15.3%的MSCs细胞可表达视紫红质。
结论 经EGF诱导后,15.3%的大鼠MSCs可分化为光感受器细胞。 |
