LAMA1基因与病理性近视的相关性研究

目的:分析病理性近视眼及非近视眼巩膜组织中的LAMA1基因在转录水平(mRNA水平)是否存在显著性差异,探讨其与病理性近视的相关性。
方法:收集8例行抗青光眼手术的原发性开角性青光眼(POAG)患者(伴有病理性近视)、5例自愿捐献角膜的健康成年人(不伴有病理性近视)及5例行抗青光眼手术的POAG患者(不伴有病理性近视)术中切除的巩膜组织,其中由行抗青光眼手术的POAG患者(伴有病理性近视)术中切除的巩膜组织作为病理性近视组,其余两组为非病理性近视组(角膜移植供眼组和青光眼组)。收集各组巩膜组织,提取总RNA,针对LAMA1基因设计特异性引物进行RT-PCR,凝胶分析系统测定各样本β-actin和LAMA1基因PCR反应产物电泳条带灰度值,以β-actin电泳条带为参照,求出各样本LAMA1基因PCR反应产物电泳条带灰度值和β-actin电泳条带灰度值的比值(LAMA1/β-actin),继而对所得比值采用单因素方差分析法进行统计分析。
结果:各样本LAMA1/β-actin在角膜移植供眼组和青光眼组之间比较无显著性差异(P>0.01) ,而在病理性近视组明显低于非病理性近视组(角膜移植供眼组和青光眼组),两组有显著性差异 (P<0.01)。
结论:病理性近视眼的巩膜组织中LAMA1基因转录水平降低,初步判定LAMA1基因与病理性近视具有相关性。
The correlation between the LAMA1 gene and pathological myopia

Abstract
Objective: To analyze whether there is significant difference on transcriptional level(mRNA level)of the LAMA1 gene in scleral tissue between pathological myopic eyes and non-myopic eyes, then to investigate the correlation between the LAMA1 gene and pathological myopia.
Methods: To collect 8 samples from the primary open-angle glaucoma POAG patients(with pathological myopia)who accepted trabeculectomy; 5 samples from the healthy volunteers without pathological myopia, who donated their corneas; 5 samples from the POAG patients without pathological myopia, who accepted trabeculectomy. The sample was laminal slceral tissue during trabeculectomy. Among them the samples which obtained from the patients accompanied with pathological myopia were in the experimental group (EG),the others were in the control group (corneal transplantation CG and glaucoma CG. Through the methods of extracting the total RNA from each sample, designing specific primer aim directly at LAMA1 gene, then RT-PCR(reverse transcription-polymerase chain reaction),the gel analytical system measured the electrophoresis strips’ gray scale values of PCR reaction products of β-actin and LAMA1 gene. In accordance with the electrophoresis strips’ gray scale values of PCR reaction products of β-actin implored the ratios of the electrophoresis strips’ gray scale values of PCR reaction products of LAMA1 gene and β-actin (LAMA1/β-actin), then used statistics methods such as analysis of variance ANOVA .
Results: LAMA1/β-actin of the experimental group are lower than that of the control group (corneal transplantation CG and glaucoma CG)significantly, there is significant difference between two groups (P<0.01) and there is no significant difference between corneal transplantation CG and glaucoma CG (P>0.01).
Conclusion: On transcriptional level (mRNA level), LAMA1 gene which is confirmed candidate gene of pathological myopia from pathological myopic scleral tissue is lower than non-myopia, so we got the primary conclusion that there is relationship between LAMA1 gene and pathological myopia.
